The control of diseases remains a question of major importance in order to reduce pathological risks in bivalve larval rearings. For such a purpose, the knowledge of diseases, pathogens and routes of infection is required. A new species of vibrio pathogenic to P. maximus larvae, Vibrio pectenicida, was described by phenotypic and genotypic characters. It belongs to the V. splendidus group, where sorne other new pathogenic species were already found. The pathogenesis of V. pectenicida was studied by histological methods and experimental exposures in conventional and axenic larval rearings. Nowadays, cupped oyster, Crassostrea gigas, production is the main shellfish production in France. Spats cultivated on most farms are usually collected in the wild, but hatchery spats are becoming more numerous, and represent around 10% of today's cultivated spats. During the summer period, when the temperatures are higher, high mortality rates have been regularly observed in cupped in oyster C. gigas (60 to 100%) spats since 1991, both in hatcheries-nurseries and in their natural habitat. Over the last five years, these episodes have become a big concern for all the professionals. |

In the frame of MOREST project, a common biological material, resulting of a mixture of different families produced in experimental hatchery, was reared in two different environmental fields : Normandy and Charente. Concomitantly, a pool was conditioned at the Ifremer Argenton hatchery with three different algae levels : 4%, 8% and 12% of algal dry weight per oyster dry weight. Bi-parental families were produced in hatchery and tested in the field in 2001. Two sets of 5 families were constituted, selected on their high (R) and low (S) survival. These two sets were reared in Brittany from March to November 2002. Samplings were performed twice a month to obtain data on biometry, survival, reproductive cycle, biochemical composition, adenylate energy charge, hemolymph parameters (ions and defense system) and muscle strength. The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Vibrio aestuarianus strain 01/32 was previously shown to be pathogenic to Crassostrea gigas juveniles. To investigate virulence mechanisms of this pathogen, we studied the toxicity to oysters of its extracellular products (ECPs). ECPs displayed lethality to animals, with a LD50 value of 3.3 mu g/g body weight. To determine the oyster cellular immune responses induced by these ECPs, we further examined in vitro their effects on C. gigas hemocytes, using flow cytometric-based hemocyte assays. Treatment of hemolymph with ECPs caused a significant inhibition of hemocyte phagocytosis and adhesive capabilities. The strategies used by bacterial pathogens to circumvent host defense mechanisms remain largely undefined in bivalve molluscs. In this study, we investigated experimentally the interactions between the Pacific oyster (Crassostrea gigas) immune system and Vibrio aestuarianus strain 01/32, a pathogenic bacterium originally isolated from moribund oysters. Arachidonic acid (20:4n-6, ArA) and its eicosanoid metabolites have been demonstrated to be implicated in immune functions of vertebrates, fish, and insects. Thus, the aim of this study was to assess the impact of ArA supplementation on the FA composition and hemocyte parameters of oysters Crassostrea gigas. Bivalve hemocyte competence has been measured by quantifying functional characteristics, including reactive oxygen intermediate (ROI) production after activation with zymosan or phorbol myristate acetate (PMA). However, untreated oyster hemocytes also produce ROI and RNI (reactive nitrogen intermediates) after bleeding even if not stimulated by Zymosan or PMA. Extensive investigation of this parameter by flow cytometry showed that, in vitro, ROI/RNI production by untreated hemocytes maintained in seawater appeared to be independent of both bacterial burden in the serum and non-self particle phagocytosis. Extracellular products (ECPs) of the pathogenic Vibrio aestuarianus 01/32 were previously reported to display lethality in Crassostrea gigas oysters and to cause morphological changes and immunosuppression in oyster hemocytes. To identify the source of this toxicity, biochemical and genetic approaches were developed. ECP protease activity and lethality were shown to be significantly reduced following incubation with metal chelators, suggesting the involvement of a zinc metalloprotease.
